Once a bank or company has given you an estimated figure for your business loan rate, are you then able to negotiate this rate or is it concrete?

You should definitely be able to negotiate the rate. But this will depend on how good your idea is, and also how “good” you look as a potential client. If you have an excellent credit rating, already have other businesses and are generally a low risk investment then I am sure you will be able to negotiate the terms.
